Collagenisanextracellularmatrixproteinthatservesasascaffolddefiningtheshapeandmechanicalpropertiesofmanytissuesandorgansincludingskin,tendon,arterywalls,fibrocartilage,boneandteeth.Type1collagenisthemostabundantproteininmammals.CollagensaresynthesizedwithN-terminalandC-terminalpropeptidesthatarecleavedduringmaturationandsecretion.Aftercleavageofthepropeptides,themostN-terminalandC-terminalremainingsequencesareknownastelopeptides.Mutationsinthecollagen1,alpha1gene(COL1A1)areknowntocauseosteogenesisimperfecta(akabrittlebonedisease)(Byers1989).FurThermore,mutationsfoundinthefirst90residuesofthehelicalregionofalpha1collagenhavebeenimplicatedinthepreventionordelayedremovaloftheprocollagenN-propeptideleADIngtoacombinedosteogenesisimperfectaandEhlers-Danlossyndrome(EDS)phenotype(Cabraletal.,2005)
