Theantibodyispreparedfrompooledrabbitserumbyaffinitypurificationviasequentialchromatographyonphospho-anddephospho-peptideaffinitycolumns.
Theionchannelsactivatedbyglutamatearetypicallydividedintotwoclasses.ThosethataresensitivetoN-methyl-D-aspartate(NMDA)aredesignatedNMDAreceptors(NMDAR)whilethoseactivatedbyα-amino-3-hydroxy-5-methyl-4-isoxalonepropionicacid(AMPA)areknownasAMPAreceptors(AMPAR).TheAMPARarecomprisedoffourdistinctglutamatereceptorsubunitsdesignated(GluR1-4)andtheyplaykeyrolesinvirtuallyallexcitatoryneurotransmissioninthebrain(Keinänenetal.,1990;HollmannandHeinemann,1994).TheGluR1subunitiswidelyexpressedthroughoutthenervoussystem.GluR1ispotentiatedbyphosphorylationatSer831whichhasbeenshowntobemediatedbyeitherPKCorCaMkinaseII(McGlade-McCullohetal.,1993;Mammenetal.,1999;Rocheetal.,1996).Inaddition,phosphorylationofthissitehasbeenlinkedtosynapticplasticityaswellaslearningandmemory(SoderlingandDerkach,2000).
