BIOLOGicalSignificance:ExpandCollapse

Theionchannelsactivatedbyglutamatearetypicallydividedintotwoclasses.ThosethataresensitivetoN-methyl-D-aspartate(NMDA)aredesignatedNMDAreceptors(NMDAR)whilethoseactivatedbyα-amino-3-hydroxy-5-methyl-4-isoxalonepropionicacid(AMPA)areknownasAMPAreceptors(AMPAR).TheAMPARarecomprisedoffourdistinctglutamatereceptorsubunitsdesignated(GluR1-4)andtheyplaykeyrolesinvirtuallyallexcitatoryneurotransmissioninthebrain(Keinänenetal.,1990;HollmannandHeinemann,1994).TheGluR1subunitiswidelyexpressedthroughoutthenervoussystem.PhosphorylationofSer845onGluR1isthoughttobemediatedbyPKAandphosphorylationofthissiteincreasestheconductanceoftheAMPAR(Rocheetal.,1996;Bankeetal.,2000).Inaddition,phosphorylationofthissitehasbeenlinkedtosynapticplasticityaswellaslearningandmemory(Leeatal.,2003;Estebanatal.,2003).
