TroponinI(cTnI)is1of3subunits,alongwithtroponinC(TnC)andtroponinT(TnT)oftroponincomplexfoundincardiacmuscle.cTnIbindstoactininthinmyofilamentstoholdthetroponin-tropomyosincomplexinplace.PhosphorylationofcardiacisoformofTnIatserines22,23intheuniqueamino-terminalendmoleculedecreasesthecalciumsensitivityofthesarcomere,promotescalciumdissociationfromtroponinCandbyextensionenhancesratesofcross-bridgecyclinganddiastolicrelaxation(Noland,Jr.etal.,1995;Nolandetal.,1989).Inaddition,studiesusingreconstitutedfibersandmutationalanalysishaveshownthatPKCphosphorylationofTnI(largelyatSer43)inhibitstheactin-crossbridgereactionandreducestheCa++dependentactomyosinATPaserateaswellasthecalciumsensitivityofforcegeneration(Noland,Jr.andKuo,1991).PhosphorylationatThr¹⁴⁴(mediatedbyseveralPKCisoforms)reducesmaximaltensiondevelopmentandcross-bridgecyclingrates(Sumandeaetal.,2008).Importantly,changesinthephosphorylationateachofthesesiteshavebeenshowntobestage-specificwithregardtocardiacdiseaseprogression(Walkeretal.,2010).
