XIRP1isamemberoftheXinfamilyofproteins,containing16Xinrepeats.TheintercalateddiskproteinXinprotectsactinfilamentsfromdepolymerization,andisabletobundleactinfilamentstointeractwithβ-catenin,whichisthoughttostABIlizeactin-basedCytoskeletons(Choietal.,2007).TheevolutionaryemergenceoftheXinparalogsmayhaveplayedakeyroleinthedevelopmentofheartchamberswithcompleteendothelialandmyocardiallayers(Grosskurthetal.,2008).LossofmXina(mouseXinhomolog)resultsincardiachypertrophyandcardiomyopathy(Gustafson-Wagneretal.,2007).PhosphorylationofSer295isplaysakeyroleinXIRP1signaling,andthereforeincardiachypertrophy,asithasbeenshownthatacutepressure-overloadedheartsrapidlyincreasedphosphorylationattheSer295residueofXIRP1.(Changetal.,2013).
