Glyceraldehyde3-PhosphateDehydrogenase(GAPDH)isametabolicenzymeresponsIBLeforcatalyzingonestepintheglycolyticpathway,thereversibleoxidativephosphorylationofglyceraldehyde3-phosphate.BecauseGAPDHisaproteinexpressedinlargeamountsandwhichisrequiredatalltimesforimportant“housekeeping”functions,levelsofGAPDHmRNAareoftenmeasuredandusedasstandardsinstudiesofmRNAexpression.Increasingly,scientistsaremakinguseofspecificantibodiestoGAPDHincomparablestudiesoflevelsofproteinexpression.ThisantibodycanbeusedasaloADIngcontrolforwesternblottingexperiments,allowingcomparisonbetweenthelevelofthisproteinandothersinacellortissue.Apartfromaroleinglycolysis,GAPDHmayhaveotherrolessuchasintheactivationoftranscription(1).GAPDHisreportedtobindtoavarietyofotherproteins,includingtheamyloidprecursorprotein,mutationsinwhichcausesomeformsofAlzheimer’sdisease,andthepolyglutaminetractsofHuntingtin,theproteinproductaberrantformsofwhicharecausativeofHuntington’sdisease(2,3).Associationswithactinandtubulinhavealsobeenreported.Theproteinmayalsohavearoleintheregulationofapoptosis,andinterestinglymigratesfromthecytoplasmintothenucleuswhencellsbecomeapoptotic(4).
