Microtubulesare25nmdiameterproteinrodsfoundinmostkindsofeukaryoticcells.Theyarepolymerizedfromadimericsubunitmadeofoneasubunitandonebtubulinsubunit.Microtubulesareassociatedwithafamilyofproteinscalledmicrotubuleassociatedproteins(MAPs),whichincludestheproteinτ (tau)andagroupofproteinsreferredtoasMAP1,MAP2,MAP3,MAP4andMAP5(Kindler&Gardner1994).MAP2ismadeupoftwo~280kDaapparentmolecularweightbandsreferredtoasMAP2aandMAP2b.Athirdlowermolecularweightform,usuallycalledMAP2c,correspondstoapairofproteinbandsrunningat~70kDaonSDS-PAGEgels.AlltheseMAP2formsarederivedfromasinglegenebyalternatetranscription,andallshareaC-terminalsequencewhichincludeseitherthreeorfourmicrotubulebindingpeptidesequences,whichareverysimilartothosefoundintherelatedmicrotubulebindingproteinτ (tau).MAP2isoformsareexpressedonlyinneuronalcellsandspecificallyintheperikaryaanddendritesofthesecells.MAP2hasbeenrecentlyshowntobethespecificreceptorfortheneurosteroidpregnenolone(Fontaine-LenoreV.etal.,2006).
