TheDENN(differentiallyexpressedinnormalandneoplasticcells)domain(DENND)isapoorlycharacterizedproteinmoduleconservedthroughoutevolution(McPhersonetal.,2011).ProteinsbearingaDENNdomainhaverecentlyemergedasthelargestfamilyofRabGEFs.AmongtheseDENNdomainproteins,DENND3isaGEFforRab12;promotingtheexchangeofGDPtoGTP,convertinginactiveGDP-boundRAB12intoitsactiveGTP-boundform(McPhersonandXu,2015;Matsuietal.,2011).ULK-mediatedphosphorylationofDENND3atserines554and572upregulatesitsGEFactivitytowardthesmallGTPaseRab12(McPhersonandXu,2015).
