Tryptophanhydroxylase(TPH)catalyzesthefirststepinthebiosynthesisofSEROtoninandmelatonin(Martinezetal.,2001).Thus,expressionofTPHcanbeusedasanindicatorofthelocalizationofserotoninandmelatonininbrain.Inmammals,serotoninbiosynthesisoccurspredominantlyinneuronswhichoriginateintheRaphenucleiofthebrain,andmelatoninsynthesistakesplacewithinthepinealgland(Haycocketal.,2002).AlthoughTPHcatalyzesthesamereactionwithintheRaphenucleiandthepinealgland,TPHactivityisrate-limitingforserotoninbutnotmelatoninbiosynthesis(Martinezetal.,2001).
