Theantibodyispreparedfrompooledrabbitserumbyaffinitypurificationusingacolumntowhichthefusionproteinimmunogenwascoupled.
Nicotinicacetylcholinereceptors(nAChRs)areionotropic,cholinergicreceptorsthataredividedinto2types;muscletypeandneuronaltype.NeuronalnAChRsarepentamericionchannelsconsistingof5identical(homopentamers)ordifferent(heteropentamers)subunits.HeteropentamericneuronalnAChRsmediatefastsynaptictransmissionintheautonomicnervoussystem.Thepredominanthetero-oligomericnAChRintheCNScontainthesubunitsα4β2,whereasα3β4prevailinthePNS.However,theexpressionofthesesubunitsvariesnotonlybyregionbutalsoduringdevelopment(Scholzeetal2011).Inthebrain,β2-containingreceptorsgreatlyoutnumberreceptorsthatcontainβ4(McGehee&Role,1995;Albuquerqueetal.,2009),andinmostbrainregions,targeteddeletionoftheβ2subunitvirtuallyabolishes[3H]-epibatidinebindingandreceptorautorADIography(Zolietal.,1998)duetotheabsenceofaβsubunitrequiredtoformfunctionalnAChRs(Champtiaux&Changeux,2004).
