KCC2iswidelythoughttobeexpressedexclusivelyinneuronswhereitisresponsIBLeformaintaininglowintracellularchlorideconcentrationtodrivehyperpolarizingpost-synapticresponsestotheinhibitoryneurotransmittersGABAandglycine.Serine940onKCC2hasbeenshowntobephosphorylatedbyPKCandhasfurtherbeendemonstratedtobethemajorsiteforPKC-dependentphosphorylationforfulllengthKCC2moleculeswhenexpressedinHEK-293cellsasphosphorylationofSer940increasedthecellsurfacestABIlityofKCC2inthissystembydecreasingit’srateofinternalizationfromtheplasmamembrane(Leeetal.,2007).
